PENGEMBANGAN DAN VALIDASI METODE ANALISIS KANDUNGAN FORMALIN DALAM BUNGA KOL YANG DIJUAL DI PASAR JAKARTA UTARA

Abstract

Penggunaan formalin sebagai pengawet makanan dilarang karena bersifat karsinogen. Kondisi lingkungan di Jakarta Utara yang bersuhu tinggi, lembab, dan berlokasi dekat laut seringkali menyebabkan penyalahgunaan formalin sebagai pengawet pada sayur-sayuran untuk meningkatkan waktu penyimpanannya, termasuk dalam bunga kol (Brassica oleracea). Penelitian ini bertujuan untuk mendapatkan metode analisis yang valid untuk penetapan formalin dalam bunga kol yang dijual di 6 kecamatan yang terletak di Jakarta Utara. Metode penelitian meliputi optimasi derivatisasi formalin, ekstraksi formalin dari bunga kol, validasi metode analisis, dan penentuan kandungan formalin dalam bunga kol, Hasil penelitian menunjukan kondisi optimum derivatisasi pada konsentrasi asam kromatropat 100 kali konsentrasi formalin, suhu derivatisasi 60°C selama 15 menit. Ekstraksi formalin dilakukan menggunakan asam fosfat 10%, kemudian sampel didestilasi. Metode memberikan hasil yang valid, pengujian spesifisitas menunjukan tidak terdapat interferensi, metode memberikan hasil yang linear pada rentang konsentrasi 2 - 10 ?g/mL. Batas deteksi dan batas kuantisasi metode berturut-turut sebesar 0,55 ?g/mL dan 1,68 ?g/mL. Metode memberikan hasil yang akurat dengan perolehan kembali sebesar 96,2%; 97,4%; dan 96,9% dan presisi dengan koefisien variasi sebesar 2,1%; 2,0%; dan 1,5% berturut-turut pada konsentrasi 4, 5, dan 6 ?g/mL. Pengujian bunga kol di 30 pasar Jakarta Utara menunjukan rerata kandungan formalin sebesar 32,42 mg per kg sampel.
